Archbishop Spalding was established in 1963 as an all-girls high school called Holy Trinity High School. The school moved to its present location in 1966 as Martin Spalding High School. It became coeducational in 1973.

In May 2003, the school acquired the adjacent Upton farm property, enlarging the school's campus to a site. The additional property insures the school's future and its ability to accommodate the academic, athletic, and extra-curricular needs of its current and future students. The addition of a new Arts and Technology wing was completed in September 2009.

A large number of Archbishop Spalding Students participate in their athletic programs due to the wide variety of the sports offered. The Cavs compete against Baltimore-area schools in the MIAA (boys) and IAAM (girls) athletic conferences. They play in either the "A" or "B" divisions of these leagues. Former Spalding athletes include current Memphis Grizzlies player Rudy Gay. Their dance team has won 1st place three years in a row at the EDA Nationals and this year will be taking a step up and competing at an even higher level at the UDA Nationals in Orlando. Cheerleading has been Nationally ranked since 1999, and has placed 1st at the MD State Championships for seven consecutive years.
